Book excerpt: E2F transcription factors were identified as the proteins responsible for transactivation of the adenovirus E2 promoter (Nevins, 1992; Slansky & Farnham, 1996) by the E1A oncoprotein. The E2F family constitutes 8 E2F genes (E2F1-8) which encodes for 10 proteins (Kong et al, 2007) and 3 DP genes (DP1, DP2/3, and DP4) (Iaquinta & Lees, 2007; Johnson & Degregori, 2006; Lavia & Jansen-Durr, 1999). E2F1 was the first to be cloned and other family members were cloned through homology to E2F1 or through binding to Rb-related proteins (DeGregori & Jolmson, 2006). The DNA binding domain of ten E2F proteins share homology with the two dimerization proteins (DP1 and DP2) (Kong et al, 2007). The central consensus sequence c/gGCGCg/c of the DNA binding site is symmetric among all the E2F family members (E2F and DP proteins) (Zheng et al, 1999). The E2F DP heterodimers bind through the consensus c/gGCGCg/c E2F site (Zheng et al, 1999).
